Sample NMR Spectra:

NMR (Nuclear Magnetic Resonance) or FT-NMR technique measures RF (radio frequency) absorption by the NMR active isotope of an element, for example, the isotope of hydrogen, when placed in a high magnetic field. The isotope is ‘NMR Active’ when it possesses the property ‘SPIN’ (I) higher than zero (I > 0). In other words, the nuclear isotope must have spin angular momentum, for it to have magnetic moment and give NMR signal and to show NMR spectrum. For example, hydrogen has 3 NMR active isotopes known as Proton (H-1, SPIN=1/2), Deuterium (D, H-2, SPIN=1) and Tritium (T, H-3, SPIN=1/2) and we can measure both Proton and Deuterium NMR from a sample of liquid Water. NMR Spectra are better classified based on the NMR active nucleus (see NMR Periodic Table) that is detected after tuning the detection coil in the probe for the maximum absorption of radio frequency (RF) radiation appropriate for that isotope in the magnetic field of the superconducting magnet of the NMR spectrometer. Each NMR active Nuclear Isotope absorbs RF radiation at different radio frequency with different intensity based mainly on its magnetic property and natural abundance unless the sample is isotopically enriched with the desired isotope. NMR spectra and its sensitivity or signal strength, therefore, varies with the nuclear isotope measured. For example, it is not uncommon to measure Nitrogen NMR from the naturally abundant N-15 isotope of a compound, such as ‘Formamide’. However, in common practice, nitrogen NMR of most Proteins, DNA and RNA samples are measured after enriching the samples with N-15 isotopes…
